چکیده

MicroRNAs (miRNAs) play important roles in post-transcriptional gene silence by directing target mRNA cleavage or translational inhibition. Currently, hundreds of miRNAs have been identified in plants, but no report has been published in wild soybean (Glycine soja). We constructed a small-RNA library consisting of 2880 sequences with high quality, in which 1347 were 19-24 nt in length. By utilizing the miRNA, Rfam and domesticated soybean EST database, we have analyzed and predicted the secondary structure of these small RNAs. As a result, 15 conserved miRNA candidates belonging to 8 different families and 9 novel miRNA candidates comprising 8 families were identified in wild soybean seedlings. All these miRNA candidates were validated by Northern blot and the novel candidates expressed in a tissue specific manner. Furthermore, putative target genes were predicted for novel miRNA candidates and two of them were verified by 5’-RACE experiments. These results provided useful information for miRNA research in wild soybean and plants.

Comparison of Small RNA Profiles of Glycine max and Glycine soja at Early Developmental Stages

Small RNAs, including microRNAs (miRNAs) and phased small interfering RNAs (phasiRNAs; from PHAS loci), play key roles in plant development. Cultivated soybean, Glycine max, contributes a great deal to food production, but, compared to its wild kin, Glycine soja, it may lose some genetic information during domestication. In this work, we analyzed the sRNA profiles of different tissues in both c...

Hybridization between GM soybean (Glycine max (L.) Merr.) and wild soybean (Glycine soja Sieb. et Zucc.) under field conditions in Japan.

Accumulation of information about natural hybridization between GM soybean (Glycine max) and wild soybean (Glycine soja) is required for risk assessment evaluation and to establish biosafety regulations in Japan. This is particularly important in areas where wild relatives of cultivated soybean are grown (i.e. East Asia including Japan). To collect information on temporal and spatial factors af...

Tracing soybean domestication history: From nucleotide to genome

Since the genome sequences of wild species may provide key information about the genetic elements involved in speciation and domestication, the undomesticated soybean (Glycine soja Sieb. and Zucc.), a wild relative of the current cultivated soybean (G. max), was sequenced. In contrast to the current hypothesis of soybean domestication, which holds that the current cultivated soybean was domesti...

Multigene family for Bowman-Birk type proteinase inhibitors of wild soja and soybean: the presence of two BBI-A genes and pseudogenes.

Genes for Bowman-Birk type protease inhibitors (BBIs) of wild soja (Glycine soja) and soybean (Glycine max) comprise a multigene family. The organization of the genes for wild soja BBIs (wBBIs) was elucidated by an analysis of their cDNAs and the corresponding genomic sequences, and compared with the counterparts in the soybean. The cDNAs encoding three types of wild soja BBIs (wBBI-A, -C, and ...

Genome Re-Sequencing of Semi-Wild Soybean Reveals a Complex Soja Population Structure and Deep Introgression

Semi-wild soybean is a unique type of soybean that retains both wild and domesticated characteristics, which provides an important intermediate type for understanding the evolution of the subgenus Soja population in the Glycine genus. In this study, a semi-wild soybean line (Maliaodou) and a wild line (Lanxi 1) collected from the lower Yangtze regions were deeply sequenced while nine other semi...
